Conventional light microscopy is constrained by the diffraction limit, which restricts resolution to about 200 nm in the x and y dimensions and 500 nm in the z dimension. Many molecules and structures of interest require higher resolution to be effectively visualized, necessitating the development of techniques that can surpass this limit.
A variety of super-resolution methods now exist, including localization-based techniques (like PALM and STORM), structural techniques (such as SIM and iSIM), and post-processing methods (like SRRF).
These approaches each have distinct advantages based on the specific sample and application, but they generally demand a scientific camera with high sensitivity, speed, and resolution. Additionally, a large field of view can provide further benefits in imaging.
